NC8 has been used as a model strain in the development of genetic tools, such as conjugation (14), transformation (2), and expression vectors (18, 19), in lactobacilli. To evaluate the efficiency of preventing pathogenic avian influenza by vaccination with recombinant Lactobacillus plantarum (L. plantarum) that expresses conserved antigens, the mucosal and systemic immune responses in animals vaccinated with recombinant L. plantarum NC8-409-1 (NC8-pSIP409-pgsA'-HA2) and NC8-409-2 (NC8-pSIP409-pgsA'-3M2e-HA2) were evaluated. To develop a more effective and safe oral vaccine against rabies, we constructed recombinant Lactobacillus plantarum NC8 carrying one or two copies of the G gene with a dendritic cell-targeting peptide (DCpep) fused at the C-terminal designated NC8-pSIP409-sRVG or NC8-pSIP409-dRVG, respectively. L. plantarumstrain NC8 (CCUG 61730) was isolated in the 1980s from grass silage and identified as naturally plasmid free, an unusual property among L. plantarumstrains.

Plantaricin NC8 (PLNC8), a coculture-inducible two-peptide bacteriocin from Lactobacillus plantarum NC8, has recently been purified and genetically characterized. Analysis of an 8.1-kb NC8 DNA region downstream of the PLNC8 operon revealed the presence of at least four operons involved in bacteriocin production, showing high homology to the plantaricin cluster in L. plantarum C11. L. plantarum strain NC8 (CCUG 61730) and the Escherichia coli- Lactobacillus shuttle vector pSIP409 were kindly provided by Professor A. Kolandaswamy (Madurai Kamaraj University, India). The pValac-GFP plasmid was kindly provided by Anderson Miyoshi [ 18 ].
